چکیده

This study was undertaken to identify an opsonin in the coelomic fluid of Strongylocentrotus nudus by separating several humoral defense factors using galactose-affinity chromatography. A galactose-binding-protein had no opsonic activity against rabbit and sheep erythrocytes, whereas the galactose-unadsorbed fraction and hemolysin fraction separated by affinity adsorption onto formaldehyde-fixed ghosts of sheep erythrocytes possessed significant opsonic activity against both erythrocytes. The opsonic and hemolytic activities of the hemolysin fraction were inactivated at the same temperature (50•Ž) at which the opsonic acitivity of the fresh coelomic fluid was lost. The opsonic activity of the hemolysin fraction was inhibited by complement inhibitors such as hydrazine and threonine. These results suggest that a hemolysin in the coelomic fluid mainly plays an important role as an opsonin in the initial internal defense system.

An Alkaline Phosphatase Lacking Wheat Germ Agglutinin Binding Sites Useful Enzyme for Lectin Assays with Comparable Activity to the Calf Enzyme

Despite the availability of various alkaline phosphatase (ALP) isoenzymes, the calf enzyme is being used in current enzyme assays as the detector enzyme. The glycosylation pattern of this enzyme makes it a suitable ligand for binding to wheat germ agglutinin lectin (WGA). As a result of this property, the enzyme can not be used as a conjugate with this lectin, and the calf enzyme conjugates can...

Innate immunity in insects: the role of multiple, endogenous serum lectins in the recognition of foreign invaders in the cockroach, Blaberus discoidalis.

Unlike vertebrates, insects do not have an Ab-based nonself recognition system, and must rely totally on innate immunity to defend themselves from microbial invaders. The most likely candidates for recognizing foreign material in insects are the lectins, which have already been shown to be important in mammalian innate immunity. The hemolymph of the cockroach, Blaberus discoidalis, contains mul...

Fluorescence Imaging of Streptococcus pneumoniae with the Helix pomatia agglutinin (HPA) As a Potential, Rapid Diagnostic Tool

Streptococcus pneumoniae is a common human pathogen and a major causal agent of life-threatening infections that can either be respiratory or non-respiratory. It is well known that the Helix pomatia (edible snail) agglutinin (HPA) lectin shows specificity for terminal αGalNAc residues present, among other locations, in the Forssman pentasaccharide (αGalNAc1→3βGalNAc1→3αGal1→4βGal1→4βGlc). Based...

Recent advances on nano delivery of Helix mucus pharmacologically active components

Bioactive products from snail slime of “Helix” specie have potential applications in preventing and/or treating several human diseases and in cancer diagnosis. However, the poor pharmacokinetics characteristics of these natural compounds limit their use. Nanotechnology offers promising solutions for the enhanced formulation of these molecules through the synthesis of nanosized drug del...

Interaction of Receptors on Lectins with Membrane Erythrocyte Surfaces

The interactions of human genotype AO erythrocytes (red blood cells) (RBCs) with N-acetylgalactosamine-reactive lectins isolated from Helix pomatia (HPA)and from Dolichos biflorus (DBA) were studied. Binding curves obtained with the use of tritium-labeled lectins showed that the maximal numbers of lectin molecules capable of binding to human genotype AO RBCs were 3.8 x 105 and 2.7 x 105 molecul...
